Background:

  • Cancer-associated thrombosis (CAT) is a significant cause of mortality in cancer patients.
  • Low-molecular-weight heparins (LMWHs), such as tinzaparin, are crucial for managing CAT.
  • Limited data exist on LMWH pharmacokinetics (PK) in cancer patients, hindering optimal treatment.

Purpose of the Study:

  • To compare tinzaparin PK in cancer versus non-cancer patients.
  • To develop a population PK model for tinzaparin.
  • To assess the influence of cancer status on tinzaparin PK.

Main Methods:

  • Prospective, multicenter, case-control trial.
  • Enrollment of patients receiving once-daily subcutaneous tinzaparin (175 IU·kg⁻¹).
  • Plasma anti-Xa activity measurement and non-linear mixed-effects modeling analysis.

Main Results:

  • A monocompartmental model with first-order absorption described tinzaparin PK.
  • Volume of distribution correlated with body weight; clearance and anti-Xa activity with creatinine clearance.

  • Cancer status did not significantly alter tinzaparin PK.
  • Further research is needed to elucidate the role of renal function in tinzaparin dosing.
