What is Gene Expression?01:42

What is Gene Expression?

194.1K
Overview
Gene expression is the process in which DNA directs the synthesis of functional products, that is, proteins. Cells can regulate gene expression at various stages. It allows organisms to generate different cell types and enables cells to adapt to internal and external factors.
Genetic Information Flows from DNA to RNA to Protein
A gene is a stretch of DNA that serves as the blueprint for functional RNAs and proteins. Since DNA is made up of nucleotides and proteins consist of amino...

What is Gene Expression?

10.8K
A gene is a stretch of DNA that serves as the blueprint for functional RNAs and proteins. Since DNA is comprised  of nucleotides and proteins are comprised of amino acids, a mediator is required to convert the information encoded in DNA into proteins. This mediator is the messenger RNA (mRNA). mRNA copies the blueprint from DNA by a process called transcription. In eukaryotes, transcription occurs in the nucleus by complementary base-pairing with the DNA template. The mRNA is then...

Regulation of Expression at Multiple Steps01:23

Regulation of Expression at Multiple Steps

1.3K
The gene expression in cells is regulated at different stages: (i) transcription, (ii) RNA processing, (iii) RNA localization, and (iv) translation. Transcriptional regulation is mediated by regulatory proteins such as transcription factors, activators, or repressors—these control gene expression by initiating or inhibiting the transcription of genes. Once a precursor or pre-mRNA is produced, it undergoes post-transcriptional modification, including 5' capping, splicing, and the...

Transcription Attenuation in Prokaryotes02:42

Transcription Attenuation in Prokaryotes

18.1K
Transcriptional attenuation occurs when RNA transcription is prematurely terminated due to the formation of a terminator mRNA hairpin structure.  Bacteria use these hairpins to regulate the transcription process and control the synthesis of several amino acids including histidine, lysine, threonine, and phenylalanine. Transcription attenuation takes place in the non-coding regions of mRNA.
There are several different mechanisms used to attenuate transcription. In ribosome mediated...

Satellite Stem Cells and Muscular Dystrophy

2.3K
Satellite stem cells or myosatellite cells are quiescent stem cells that Alexander Mauro first identified in 1961. These cells are located between the sarcolemma, the plasma membrane of muscle fibers, and the basal lamina, the connective tissue sheath covering it. These mononucleated cells are activated in response to muscle injury, can transform into myoblasts, and may form or repair muscle fibers. Myosatellite cells can provide additional myonuclei for muscle regeneration or return to a...

MicroRNAs01:22

MicroRNAs

23.9K
MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After...

Area of Science:

  • Genetics
  • Molecular Biology
  • Epigenetics

Background:

  • Satellite DNAs are abundant genomic sequences vital for centromere and heterochromatin structure.
  • While their role in gene regulation is suggested, the underlying molecular mechanisms remain unclear.

Purpose of the Study:

  • To investigate the role of satellite transcripts in gene expression regulation using human alpha satellite as a model.
  • To elucidate the molecular mechanisms by which satellite RNA influences gene transcription.

Main Methods:

  • Generation of cell lines with forced, exogenous overexpression of alpha satellite RNA.
  • Analysis of expression levels of genes containing alpha satellite repeats within introns.
  • Assessment of histone modifications at intronic alpha satellite loci.

Main Results:

  • A positive correlation was observed between exogenous alpha satellite RNA expression and the downregulation of alpha-associated genes.
  • Elevated alpha satellite RNA levels did not alter key histone modifications associated with pericentromeric heterochromatin or euchromatin.
  • Evidence suggests alpha satellite RNA forms RNA-DNA hybrids at intronic satellite loci, potentially affecting chromatin and transcription.

Conclusions:

  • Alpha satellite RNA plays a novel role in modulating the expression of associated genes, beyond its known functions in centromere and heterochromatin assembly.
  • RNA-DNA hybrid formation at intronic satellite loci is a proposed mechanism for alpha satellite RNA-mediated gene regulation.
