Background:

  • Sirtuins are a class of proteins involved in various cellular processes, including DNA repair, metabolism, and aging.
  • Dysregulation of sirtuin expression has been implicated in the development and progression of various cancers.
  • Sirtuin 1 (SIRT1) and sirtuin 2 (SIRT2) are key members of this family, with emerging roles in cancer biology.

Purpose of the Study:

  • To investigate the expression levels of SIRT1 and SIRT2 in colorectal cancer (CRC) tissues.
  • To evaluate the association between SIRT1 and SIRT2 expression and clinicopathological parameters of CRC.
  • To explore the potential diagnostic and prognostic value of SIRT1 and SIRT2 in CRC.

Main Methods:

  • Immunohistochemistry was used to assess SIRT1 and SIRT2 immunoreactivity in 104 colorectal adenocarcinoma specimens.

  • SIRT2 expression is significantly correlated with advanced TNM stage and the presence of metastasis in colorectal cancer.
  • Both SIRT1 and SIRT2 show a significant association with K-RAS mutations, indicating their involvement in the molecular pathology of CRC.
  • These findings suggest that SIRT1 and SIRT2 may serve as potential biomarkers for colorectal cancer diagnosis and prognosis.
