Microwave-Promoted One-Pot Sustainable Synthesis of Fluorogenic Amino Acids: Late-Stage Diversification of Peptides

Abstract:
A microwave-mediated sustainable protocol is outlined herein for rapid access to pyrimidine-based fluorogenic amino acids from aryl methyl ketone, aryl aldehyde, and arginine precursors. This solvent- and transition metal-free reaction is catalyzed by sodium carbonate, providing an important route to a library of pyrimidine-based fluorogenic α-amino acids as well as peptides. The method successfully demonstrates the late-stage diversification of peptides, providing the potential for drug discovery and molecular imaging. Notably, this approach simplifies the synthesis of fluorogenic amino acids and peptides by eliminating the need for multiple reaction steps, transition metal catalysts, organometallic reagents, and protecting group manipulations, thus minimizing waste, time, and cost.
