Background:

  • Alzheimer's disease (AD) is a leading cause of dementia, characterized by amyloid-beta (Aβ) plaques and tau tangles.
  • Extracellular vesicles (EVs) are cell-secreted nanoparticles that can transport biomolecules and influence neuropathology.
  • The specific role of EVs in AD-related behavioral deficits remains largely unexplored.

Purpose of the Study:

  • To investigate the impact of brain-derived extracellular vesicles (EVs) on memory and behavior in a mouse model.
  • To determine if EVs from Alzheimer's disease (AD) models can induce cognitive deficits.

Main Methods:

  • Extracellular vesicles (EVs) were isolated from post-mortem brain tissue of control and AD patients, as well as from APP/PS1 mice.
  • These EVs were stereotactically injected into the hippocampi of wild-type (WT) mice.
  • Cognitive function was evaluated using behavioral tests at various time points post-injection.

Main Results:

  • Mice injected with EVs from control brains showed no motor or behavioral impairments.
  • Conversely, mice receiving EVs from AD brains exhibited significant memory deficits.
  • These cognitive impairments were evident 14 and 21 days after injection, confirming AD-EVs and APP/PS1-EVs induce memory loss.

Conclusions:

  • Extracellular vesicles (EVs) can transmit AD-associated pathological molecules and induce disease-like memory impairment.
  • These findings highlight EVs as a critical factor in AD pathogenesis and progression.
  • EVs represent a promising therapeutic target for neurodegenerative diseases like Alzheimer's.
