  • Baseline neuroimaging features may predict future cognitive changes in SCD.

Purpose of the Study:

  • To determine if baseline brain amyloid-beta (Aβ) deposition, hippocampal volume, and glucose metabolism predict cognitive changes in SCD.
  • To identify imaging biomarkers for predicting SCD progression.

Main Methods:

  • 95 subjects (59 SCD, 36 controls) underwent baseline PET/CT (Aβ, glucose metabolism) and MRI (hippocampal volume).
  • Cognitive function was assessed at baseline and after two years.
  • General linear models analyzed the relationship between baseline imaging features and cognitive change rates.

Main Results:

  • SCD subjects showed a trend toward accelerated executive function decline.
  • Baseline whole-brain Aβ deposition predicted changes in naming and executive function.
  • Hippocampal volume and parahippocampal glucose metabolism predicted changes in overall cognition, naming, and executive function.

Conclusions:

  • Baseline "A" (amyloid) and "N" (neurodegeneration) imaging features predict longitudinal cognitive changes in SCD.
  • These imaging biomarkers are key for monitoring SCD progression.
  • SCD patients experience accelerated executive function decline over two years.
