Release of Pro-Inflammatory Cytokines from Macrophages Stimulated by Denture Base Materials in Smokeless Tobacco

Background:
Denture base materials, when exposed to carcinogenic substances like smokeless tobacco extracts (STE), may provoke inflammatory responses in oral tissues. Understanding macrophage-mediated cytokine release in such environments is critical to evaluating the biocompatibility of these materials.
Materials And Methods:
Three denture base materials-conventional heat-cured poly (methyl methacrylate) (PMMA), high-impact PMMA, and CAD/CAM-milled PMMA-were evaluated. RAW 264.7 macrophages were exposed to material eluates with and without STE. Cytokine levels were quantified using ELISA.
Results:
All materials showed significantly increased cytokine release in STE environments, with conventional PMMA eliciting the highest levels. CAD/CAM PMMA showed the least cytokine induction.
Conclusion:
Exposure to smokeless tobacco enhances the pro-inflammatory potential of denture base materials, with conventional PMMA being the most reactive. CAD/CAM PMMA demonstrates superior biocompatibility in inflammatory conditions.
