In Vitro Calcium Imaging in Short-Term and Long-Term Rat Hippocampal Cultures03:41

In Vitro Calcium Imaging in Short-Term and Long-Term Rat Hippocampal Cultures

Source: Calvo-Rodríguez, et al. Fluorescence and Bioluminescence Imaging of Subcellular Ca2+ in Aged Hippocampal Neurons. J. Vis. Exp. (2015). This video demonstrates the use of fluorescence imaging to measure cytosolic calcium levels in short-term and long-term rat hippocampal cultures. By comparing young and aged neurons, it highlights how increased L-type voltage-gated calcium channel density and prolonged activation in aged neurons result in calcium overloading, a...

Isolation Method for Long-Term and Short-Term Hematopoietic Stem Cells

2.5K
We present a step-by-step protocol for the isolation of long-term hematopoietic stem cells (LT-HSCs) and short-term HSCs (ST-HSCs) using the Hoxb5 reporter...

Endocarditis II: Clinical Features of Infective Endocarditis

434
Endocarditis can present various clinical features depending on the causative organism and the patient's underlying health conditions. Initially, the clinical features of infective endocarditis develop gradually, presenting with nonspecific symptoms that can be easily mistaken for other illnesses.General SymptomsEarly symptoms of infective endocarditis are fever, chills, weakness, malaise, fatigue, and weight loss. These symptoms reflect the systemic nature of the infection and the body's...

Short-Term Free-Floating Slice Cultures from the Adult Human Brain

9.6K
A protocol to prepare free-floating slice cultures from adult human brain is presented. The protocol is a variation of the widely used slice culture method using membrane inserts. It is simple, cost-effective, and recommended for running short-term assays aimed to unravel mechanisms of neurodegeneration behind age-associated brain...

Source: Lee, D. et. al., Modified Experimental Conditions for Noise-Induced Hearing Loss in Mice and Assessment of Hearing Function and Outer Hair Cell Damage. J. Vis. Exp. (2023)This video demonstrates the procedure of noise-induced hearing loss in mice. High-volume sound damages hair cells in the inner ear and disrupts auditory pathways, resulting in hearing...

Summary

Culture-negative infective endocarditis (IE) is challenging, even after antibiotics. Molecular tools like 16S rRNA PCR can identify bacteria, guiding treatment for IE when blood cultures are negative.

Area of Science:

  • Infectious Diseases
  • Microbiology
  • Cardiology

Background:

  • Blood culture-negative infective endocarditis (IE) presents significant diagnostic challenges.

  • Prior antibiotic exposure is a common reason for negative blood cultures in IE.
  • Delayed diagnosis of IE can lead to severe complications.
