Transcription

156.8K
Overview
Transcription is the process of synthesizing RNA from a DNA sequence by RNA polymerase. It is the first step in producing a protein from a gene sequence. Additionally, many other proteins and regulatory sequences are involved in the proper synthesis of messenger RNA (mRNA). Regulation of transcription is responsible for the differentiation of all the different types of cells and often for the proper cellular response to environmental signals.

Nuclear Fusion02:45

Nuclear Fusion

33.9K
The process of converting very light nuclei into heavier nuclei is also accompanied by the conversion of mass into large amounts of energy, a process called fusion. The principal source of energy in the sun is a net fusion reaction in which four hydrogen nuclei fuse and ultimately produce one helium nucleus and two positrons.
A helium nucleus has a mass that is 0.7% less than that of four hydrogen nuclei; this lost mass is converted into energy during the fusion. This reaction produces about...

Master Transcription Regulators02:23

Master Transcription Regulators

7.8K
Master transcription regulators are regulatory proteins that are predominantly responsible for regulating the expression of multiple genes. Often these genes work in concert to drive a  complex process. Activation of a master transcription regulator can lead to a cascade of transcriptional activation necessary for that outcome. These regulators can directly bind to the regulatory sequences of the various genes involved, or they can indirectly regulate transcription by binding to regulatory...

Eukaryotic Transcription Inhibitors01:52

Eukaryotic Transcription Inhibitors

11.0K
Certain biochemical processes, such as embryonic development and cell growth regulation, depend on the repression of specific genes. DNA binding proteins known as eukaryotic transcription inhibitors regulate the repression of gene expression in eukaryotes. The presence of these inhibitors at the required location and time in the cell is triggered by the presence of hormones and additional signals from other cells.
Eukaryotic transcription inhibitors usually contain two distinct domains, a...

Uncertainty in Measurement: Reading Instruments

Counting is the type of measurement that is free from uncertainty, provided the number of objects being counted does not change during the process. Such measurements result in exact numbers. By counting the eggs in a carton, for instance, one can determine exactly how many eggs are there in the carton. Similarly, the numbers of defined quantities are also exact. For example, 1 foot is exactly 12 inches, 1 inch is exactly 2.54 centimeters, and 1 gram is exactly 0.001 kilograms. Quantities...

Transcription Elongation Factors02:35

Transcription Elongation Factors

14.0K
Transcription elongation is a dynamic process that alters depending upon the sequence heterogeneity of the DNA being transcribed. Hence, it is not surprising that the elongation complex's composition also varies along the way while transcribing a gene.
The transcription elongation is regulated via pausing of RNA polymerase on several occasions during transcription. In bacteria, these halts are necessary because the transcription of DNA into mRNA is coupled to the translation of that mRNA...

Area of Science:

  • Genomics
  • Transcriptomics
  • Cancer Biology

Background:

  • Fusion transcripts, arising from distinct genes, are crucial for identifying cancer biomarkers and therapeutic targets.
  • Detecting all fusion transcripts at the RNA level is vital for cancer research.

Purpose of the Study:

  • To identify and characterize fusion transcripts in Mantle Cell Lymphoma (MCL) using long-read RNA sequencing.
  • To investigate novel fusion transcripts, including those involving long non-coding RNAs.

Main Methods:

  • Employed Iso Sequencing (third-generation long-read RNA sequencing) on MCL cell lines.
  • Validated identified fusion transcripts using orthogonal techniques.
  • Investigated the effect of CPSF73/CPSF3 inhibition on a specific fusion transcript.

Main Results:

  • Revealed extensive transcript diversity and numerous novel transcripts in MCL.
  • Identified several novel fusion transcripts, some of which were among the longest in the MCL transcriptome.
  • Validated known (CTBS::GNG5) and novel fusion transcripts (RBM15::LAMTOR5:AS), including one involving a long non-coding RNA.
  • Observed increased expression of RBM15::LAMTOR5:AS upon treatment with JT-607, a CPSF73/CPSF3 inhibitor.

Conclusions:

  • The study identified a novel fusion transcript (RBM15::LAMTOR5:AS) and numerous other fusion transcripts in MCL.
  • These findings underscore the importance of unbiased approaches for comprehensive fusion transcript identification.
  • The discovered fusion transcripts contribute to understanding transcriptome complexity in cancer and normal biology.
