Detection of stipe rot disease in Morchella sextelata based on enhanced Fourier transform infrared spectroscopy

Abstract:
Timely detection of stipe rot disease is crucial for ensuring the yield and quality of Morchella sextelata (morel). In this study, we propose a volatile-enhanced Fourier transform infrared (FTIR) spectroscopy method for detecting stipe rot disease. The signature volatiles of disease were analyzed and identified by long-path enhanced FTIR spectroscopy. Moreover, the enrichment-enhanced spectral properties of the signature volatile were analyzed using metal-organic framework (MOF) enrichment-enhanced FTIR spectroscopy. A multivariate quantitative model and an identification model for the stipe rot disease were established based on enrichment-enhanced spectral characteristics combined with chemometric methods. The results show that ammonia is the signature volatile, and the quantitative model exhibits a superior ability to quantify ammonia. The support vector machine enables the identification of disease severity and achieves a recognition accuracy of 98.75%. The volatile-enhanced FTIR technique provides a feasible method for the rapid and non-destructive detection of stipe rot disease in morel.
