    Summary

    Platelet-activating factor (PAF) from rat macrophages aggregates platelets and releases vasoactive amines. This study identifies a novel inflammatory role for macrophages in mediating these effects.

    Background:

    • Platelet-activating factor (PAF) is a key mediator in anaphylaxis.
    • PAF release from human and rabbit basophils causes platelet aggregation and vasoactive amine release.

    Purpose of the Study:

    • To investigate the cellular source and function of PAF in rat peritoneal cells.
    • To elucidate the role of macrophages in PAF-mediated inflammatory responses.

    Main Methods:

    • Rat peritoneal cells (PC) were stimulated with ionophore A23187 to induce PAF release.
    • PC were fractionated using 5-15% Ficoll gradients.
    • PAF presence was assessed in different cell fractions.

    Main Results:

    • PAF was primarily isolated from macrophage-rich fractions of PC.
    • PAF was predominantly found in adherent cell populations, not non-adherent cells.
    • Mastocyte-rich fractions did not yield significant PAF.

    Conclusions:

    • Macrophages play a significant role in the release of Platelet-Activating Factor (PAF).
    • This study suggests a novel function for macrophages in platelet aggregation and the release of inflammatory mediators.
    • Macrophage-derived PAF contributes to inflammatory processes.
