Herein, we develop a metal-free catalytic method for synthesizing sulfur-containing peptide derivatives through the reaction of dehydroalanine peptides with thiols under mild and simple conditions. This hydrothiolation enables selective construction of various C-S bonds containing sensitive groups with certain compounds being obtained in nearly stoichiometric yields. The methodology demonstrates broad functional group tolerance and high selectivity, allowing for the preparation of monofunctionalized organosulfur compounds.
