Stereospecific N-Glycosylation Strategy for β-N-Glycoside Synthesis via Base Catalysis

Conventional N-glycosylation methods predominately rely on acidic conditions, facing the challenges of low stereoselectivity and limited substrate applicability. Herein, we present a versatile and highly β-stereoselective N-glycosylation of diverse N-aglycones with easily accessible 2-nitroglycal donors via base catalysis under mild conditions, enabling facile access to a broad spectrum of β-N-glycosides bearing heteroaryl, aryl, alkyl, and nucleobase functionalities. This approach features high yields and outstanding β-stereocontrol and obviates the need for traditional stereodirecting groups. Notably, late-stage modification of structurally complex drug molecules demonstrates the synthetic potential of this powerful approach.
