We demonstrate a low-cost and low-effort method to accurately quantify bacterial concentrations in milk spanning 4 log10 CFUs, which requires just one dilution followed by micro-pipetting 3 sets (100 μL, 10 μL, 1 μL) of 5 aliquots each into 15 micro-wells (MPN approach). Good correlation (R2 > 0.9) is obtained with pour-plate methods that require significantly more effort.
