Abstract:
The 3,6-dihydro-1,2-oxazine scaffold represents a privileged structure in bioactive molecules. Herein, we report a copper-catalyzed multicomponent reaction (MCR) strategy that orchestrates the convergent [4 + 1 + 1] annulation to construct such heterocycles, from 1,3-dienes, dioxazolones, and H2O. Distinct from established copper-nitrene reactivity, this protocol steers the reactive intermediate toward a three-component cyclization, circumventing the pitfall of 2 + 1 aziridination and X-H insertion. Water acts as the sole structural oxygen source, effectively addressing the challenge of utilizing simple heteroatom sources (H2O) to build molecular complexity. This method features mild conditions, exclusive regioselectivity, and broad functional group compatibility, offering a modular alternative to traditional nitroso Diels-Alder (NDA) chemistry.

Regioselectivity and Stereochemistry of Hydroboration
Hydroboration proceeds in a concerted fashion with the attack of borane on the π bond, giving a cyclic four-centered transition state. The –BH2 group is bonded to the less substituted carbon and –H to the more substituted carbon. The concerted nature requires the simultaneous addition of –H and –BH2 across the same face of the alkene giving syn stereochemistry.
Preparation of Epoxides
Epoxides result from alkene oxidation, which can be achieved by a) air, b) peroxy acids, c) hypochlorous acids, and d) halohydrin cyclization.
Epoxidation with Peroxy Acids
Epoxidation of alkenes via oxidation with peroxy acids involves the conversion of a carbon–carbon double bond to an epoxide using the oxidizing agent meta-chloroperoxybenzoic acid, commonly known as MCPBA. Since the O–O bond of peroxy acids is very weak, the addition of electrophilic oxygen of peroxy acids to...
