Abstract:
Pseudomonas aeruginosa is a clinically common multidrug-resistant pathogen that causes infections with high mortality and great treatment difficulty. This study achieved two key innovations: precise α/β configuration control of d-GalpN3, d-GlcpN3, d-QuipN3, and d-FucpN3 sugar-linker compounds and an innovative one-pot bis-glycosylation strategy, successfully completing the total synthesis of its LPS outer core pentasaccharide (a key vaccine antigen). This work provides a promotable methodological framework for the precise synthesis of complex bacterial oligosaccharides.
