Simultaneous Determination of Capmatinib and Resveratrol via Micellar-Enhanced Synchronized Fluorescence

Abstract:
A novel, eco-friendly, and highly sensitive synchronous spectrofluorimetric method was developed and validated for the first time to simultaneously determine the synergistic chemotherapy agents, capmatinib (CMT) and resveratrol (RES). The method relies on the native fluorescence characteristics of both drugs, ensuring excellent sensitivity and selectivity without prederivatization. Simultaneous quantification was achieved at 407 nm for CMT and 306 nm for RES with a wavelength difference (∆λ) of 80 nm, using water and 1.5 mL of 1% Kolliphor as a green solvent system. Validation was performed in accordance with ICH guidelines, confirming satisfactory linearity, precision, accuracy, and selectivity. The linear concentration ranges were 50.0-1800.0 and 5.0-100.0 ng/mL for CMT and RES, with detection limits of 11.69 and 1.34 ng/mL, respectively. Owing to its high sensitivity, the developed method was successfully applied to human plasma samples, achieving excellent recovery values of 95.04%-105.54% and 94.60%-104.52% for CMT and RES, respectively, which supports its bioanalytical suitability. Moreover, the method was applied for trace analysis of both drugs in tap water and river water samples with %recoveries of 95.03%-99.14% and 94.34%-101.08% for CMT and RES, respectively. The environmental sustainability along with the operational efficiency of the proposed approach was further assessed.
