Abstract:
This study establishes a streamlined one-step strategy using optimized episomal plasmid reprogramming to directly generate extended pluripotent stem cells (dEPSCs) from human peripheral blood mononuclear cells (PBMCs), bypassing the induced pluripotent stem cell(iPSCs) intermediary phase and reducing 50% of the generation compared to conventional protocols. Functional analyses confirm that dEPSCs (direct extended pluripotent stem cells), exhibit genomic stability, bidirectional developmental potential, and shared similar molecular characteristics with traditional extended pluripotent stem cells (EPSCs), Collectively, this strategy provides a robust and scalable platform for pluripotent stem cell research, with potential applications in disease modeling, lineage-specific differentiation, and regenerative medicine.
