Background:

  • Protein secretion is vital for immune homeostasis but its molecular mechanisms are not fully understood.
  • Subcellular interactions governing protein trafficking are less explored than transcriptional regulation.

Purpose of the Study:

  • To systematically map protein-protein interactions (PPIs) in interleukin-2 (IL-2) secretion from human T cells.
  • To identify novel molecular components involved in the cytokine secretion pathway.

Main Methods:

  • Proximity-based labeling coupled with mass spectrometry was used to map PPIs.
  • Time-resolved profiling of PPIs and transcriptomic changes after T-cell stimulation was performed.

Main Results:

  • Significant enrichment of proteins involved in conventional secretory pathways (ER-to-Golgi transport, protein folding, vesicle trafficking) was identified.
  • Several identified proteins were validated as critical for efficient IL-2 secretion.
