Summary

Quick, Easy, Cheap, Effective, Rugged, Safe (QuEChERS) methods coupled with LC-MS/MS or HRMS are effective for analyzing persistent per- and polyfluoroalkyl substances (PFAS) in food. These approaches meet regulatory limits and expand analytical scope for PFAS research.

Area of Science:

  • Environmental Chemistry
  • Analytical Chemistry
  • Food Safety

Background:

  • Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants requiring sensitive analytical methods for food analysis.
  • Regulatory bodies have established strict performance criteria for methods analyzing PFAS in food matrices.
  • Existing analytical techniques face challenges in comprehensively detecting the diverse range of PFAS compounds.

Purpose of the Study:

  • To systematically review and evaluate the performance of Quick, Easy, Cheap, Effective, Rugged, Safe (QuEChERS) extraction methodologies for per- and polyfluoroalkyl substances (PFAS) determination in food.
  • To assess the suitability of QuEChERS combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) and high-resolution mass spectrometry (HRMS) for regulatory and research purposes.
  • To identify gaps and underrepresented areas in current QuEChERS-based PFAS analysis in food.

Main Methods:

  • Systematic literature review of peer-reviewed articles published between 2010 and 2025.
  • Searched Scopus, Web of Science, and Google Scholar databases up to December 18, 2025.
  • Included studies analyzing PFAS in food matrices using QuEChERS extraction with LC-MS/MS or HRMS, reporting performance/validation data.

Main Results:

  • Twenty-four studies met the inclusion criteria, predominantly using acidified acetonitrile (ACN)-based QuEChERS workflows.
  • Achieved limits of quantification (LOQ) are generally compatible with EU regulations (2023/915, 2022/1428).
  • Analytical scope expanded significantly, from 9-15 legacy PFAS to over 40 analytes, though short-chain PFAS and methods from developing countries remain underrepresented.

Conclusions:

  • QuEChERS extraction coupled with LC-MS/MS and HRMS provides a robust platform for regulatory PFAS monitoring and research in food.
  • These methods demonstrate the capability to meet stringent regulatory requirements for PFAS detection.
  • Further research is needed to address underrepresented areas, such as short-chain PFAS analysis in vegetables and methods developed in developing nations.
