    Background:

    • The mammary gland's cellular composition during immune stimulation is not fully understood.
    • Ascaris suum is a common parasite that can elicit immune responses.

    Purpose of the Study:

    • To investigate the cellular changes in the bovine mammary gland following stimulation with Ascaris suum extract.
    • To characterize the immune cells, particularly eosinophils, recruited to the mammary gland.

    Main Methods:

    • Infusion of crude Ascaris suum extract into the teat canal of heifers.
    • Assessment of mammary gland cells over two weeks post-stimulation.
    • Isolation and purification of eosinophils using Ficoll-Hypaque flotation and column filtration.
    • Surface marker analysis (Fc and complement receptors) on purified eosinophils.

    Main Results:

    • Mammary glands initially showed few cells, mainly macrophages.
    • Within 12 hours, there was a significant influx of eosinophils and neutrophils.
    • Eosinophils constituted approximately 50% of the stimulated cell population.
    • Purified eosinophils exceeded 90% purity and expressed Fc and complement receptors.

    Conclusions:

    • Ascaris suum extract effectively induces an inflammatory response in the bovine mammary gland, characterized by eosinophil and neutrophil recruitment.
    • Eosinophils play a significant role in this immune response and express key receptors for immune complex binding.
