Summary

Cold stratification overcomes seed dormancy by promoting gibberellin (GA) biosynthesis, which enhances alpha-amylase expression for germination. It also reduces abscisic acid (ABA) inhibition without affecting seedling development.

Area of Science:

  • Plant Biology
  • Molecular Biology
  • Seed Physiology

Background:

  • Seed dormancy and germination are crucial for plant establishment.
  • Cold stratification is a common method to break seed dormancy.
  • The molecular mechanisms underlying stratification's effects are not fully understood.

Purpose of the Study:

  • To investigate the molecular mechanisms by which cold stratification overcomes seed dormancy in Arabidopsis thaliana.
  • To determine the role of gibberellin (GA) and abscisic acid (ABA) signaling in stratification-induced germination.

Main Methods:

  • Arabidopsis thaliana mutants with altered GA or ABA pathways were used.
  • Seeds were subjected to cold stratification (4°C).
  • Alpha-amylase expression, enzymatic activity, and key gene expression (RGL2, ABI5) were analyzed.

Main Results:

  • Stratification broke dormancy in wild-type and ABA-related mutants but not in GA-biosynthesis mutants.
  • Stratification increased alpha-amylase transcript and activity, promoting starch hydrolysis.
  • Stratification suppressed RGL2 and ABI5 expression, indicating altered GA and ABA signaling.

Conclusions:

  • Stratification initiates GA biosynthesis, overcoming RGL2-mediated repression of alpha-amylase.
  • Stratification mitigates ABA inhibition of germination but does not affect ABA's role in post-germination development.
