Tumor Immunotherapy01:27

Tumor Immunotherapy

Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.
Combination Therapies and Personalized Medicine02:50

Combination Therapies and Personalized Medicine

Combining two or more treatment methods increases the life span of cancer patients while reducing damage to vital organs or tissue from the overuse of a single treatment. Combination therapy also targets different cancer-inducing pathways, thus reducing the chances of developing resistance to treatment.
The combination of the drug acetazolamide and sulforaphane is a good example of combination therapy to treat cancer. The cells in the interior of a large tumor often die due to the hypoxic and...

Area of Science:

  • Oncology
  • Immunotherapy
  • Radiochemistry

Background:

  • Metastatic cancers pose significant treatment challenges.
  • Radiopharmaceutical therapy (RPT) and immune checkpoint inhibitors (ICIs) are advanced cancer treatments.
  • RPT targets tumors with radiation and modulates the tumor microenvironment (TME).

Purpose of the Study:

  • To review clinical trials combining RPT and ICIs for metastatic cancers.
  • To evaluate the therapeutic synergy and clinical outcomes of RPT-ICI combination therapy.
  • To identify challenges and future directions for RPT-ICI treatment.

Main Methods:

  • Literature review of clinical trials investigating RPT and ICI combinations.
  • Analysis of preclinical data supporting RPT-ICI synergy.
  • Evaluation of reported patient outcomes, toxicity profiles, and TME modulation.

Main Results:

  • Emerging clinical trials demonstrate therapeutic synergy between RPT and ICIs.
  • Combination therapy shows potential for improved metastatic cancer outcomes.
  • Challenges include managing toxicity and overcoming immunosuppressive TME.

Conclusions:

  • RPT combined with ICIs offers a promising approach to enhance antitumor immunity.
  • Addressing challenges is crucial for optimizing RPT-ICI clinical application.
  • This combination therapy has the potential to redefine metastatic cancer care.
