ARF beyond p53: Shaping the Pancreatic Tumor Microenvironment

Abstract:
ARF (p14ARF) is a tumor suppressor classically known for stabilizing and activating p53. In this issue of Cancer Research, Ferreira and colleagues provide insights into the role of ARF in the development of pancreatic ductal adenocarcinoma, showing its p53-independent tumor microenvironment regulatory function. The findings reveal a significant association of ARF loss with stromal remodeling characterized by increased extracellular matrix deposition and fibroblast infiltration. Although further studies are needed to explore the underlying mechanisms and to determine clinical significance, this study has highlighted ARF as a potential upstream regulator of the microenvironment and a candidate therapeutic target.
