A new rapid multiplex assay simultaneously detects foodborne viruses like norovirus and hepatitis A. This portable system offers quick, on-site detection, improving public health safety.

Area of Science:

  • Foodborne Pathogen Detection
  • Molecular Diagnostics
  • Public Health Microbiology

Background:

  • Human norovirus (HuNoV) and hepatitis A virus (HAV) are major foodborne pathogens causing widespread illness.
  • Current detection methods (RT-qPCR, RT-ddPCR) are sensitive but slow and require lab equipment, hindering on-site testing.

Purpose of the Study:

  • To develop a rapid, simultaneous detection method for HuNoV (GI, GII) and HAV.
  • To create a portable molecular diagnostic platform for foodborne virus surveillance.

Main Methods:

  • Developed a multiplex reverse transcription recombinase polymerase amplification (RT-RPA) assay.
  • Integrated CRISPR-Cas12a and lateral flow detection for simultaneous identification.
  • Optimized primers and crRNAs for broad genotype coverage and specificity.

Main Results:

  • Achieved limits of detection: HAV (10¹ copies/µL), GII HuNoV (10² copies/µL), GI HuNoV (10³ copies/µL).
  • Total assay time from RNA to result was 50 minutes.
  • Demonstrated no cross-reactivity with other common foodborne viruses; validated in spiked shellfish samples.

Conclusions:

  • The multiplex RT-RPA-CRISPR-Cas12a assay provides rapid, simultaneous detection of HuNoV and HAV.
  • This portable system is a promising tool for on-site foodborne virus detection and public health monitoring.
