Abstract:
Constructing overexpression vectors for genes is a common method for studying gene function. Similarly, the open reading frame (ORF) of chimeric RNA can be cloned into eukaryotic expression vectors to explore the functions of chimeric RNAs. Here, taking the chimeric RNA BCL2L2-PABPN1 as an example, we provide a detailed protocol for inserting the BCL2L2-PABPN1 fusion ORF into a mammalian expression vector. This approach enables the overexpression of the BCL2L2-PABPN1 fusion protein in a bladder cancer cell line through retroviral or lentiviral transduction.
