We developed ARGpredict, a web server to predict protein arginylation, a crucial post-translational modification. This tool aids in understanding cellular development and stress adaptation by identifying arginylation sites.
Area of Science:
- Biochemistry
- Molecular Biology
- Bioinformatics
Background:
- Arginylation is a conserved post-translational modification (PTM) where arginine is attached to specific residues like aspartic acid, glutamic acid, and oxidized cysteine.
- This modification is catalyzed by arginyltransferase 1 (ATE1) using arginyl-tRNA and occurs at protein N-termini and mid-chain.
- Arginylation plays vital roles in cellular development, cytoskeletal organization, and adaptation to cellular stress.
Purpose of the Study:
- To develop a computational tool for predicting arginylation events in protein sequences.
- To provide an alternative to time-consuming laboratory-based experimental procedures for identifying arginylation likelihood.
Main Methods:
- Development of the ARGpredict web server.
- Implementation of algorithms to analyze protein sequences and identify potential arginylation target sites.
- Scoring of identified sites to predict arginylation likelihood.
Main Results:
- The ARGpredict web server was created to predict arginylation events.
- The server analyzes protein sequences, identifies target sites, and assigns prediction scores.
- Validation against known and experimentally verified datasets demonstrated promising prediction accuracy, supported by statistical analysis.
Conclusions:
- ARGpredict offers an effective computational approach for predicting protein arginylation.
- The web server facilitates research into the biological roles of arginylation in cellular processes.
- ARGpredict is accessible online for researchers at https://iitg.ac.in/sachinku/ARGpredict/.
