Datopotamab deruxtecan in solid tumors: Recent research progress and clinical applications

Following the approval of the first antibody-drug conjugate (ADC) for solid tumors, the therapeutic landscape in cancer has been revolutionized. Datopotamab deruxtecan (Dato-DXd) is a TROP2-targeting ADC which is composed of a humanized monoclonal antibody and a potent topoisomerase I inhibitor via a tetrapeptide-based cleavable linker. Based on data from multiple clinical trials, Dato-DXd has demonstrated compelling efficacy and a manageable safety profile. Herein, we provide an overview of recent published clinical trial data on Dato-DXd across various solid tumors. In addition, we propose future research directions to address critical issues in the clinical application of Dato-DXd, including optimal sequencing strategies with other TROP2-targeting ADCs, approaches to overcome drug resistance, and management of treatment-related adverse events.
