Abstract:
We isolated a novel strain GXU_MW_L88T from coral Porites sp. Strain GXU_MW_L88T was Gram-stain-negative, aerobic, non-spore-forming, short rod, with a capacity to produce vitamin B12. The growth requirements of this strain were a temperature range of 20-37 °C, a pH range of 5.0-11.0 and a NaCl concentration of 0-6% (w/v). Phylogenetic analysis based on 16S rRNA gene sequence showed that GXU_MW_L88T shared an identity of 92.2% with Lutimaribacter marinistellae SF-12T. The highest maximum average nucleotide identity was 72.13% to Paracoccus sediminicola SCSIO 76264T, while the highest average amino acid identity was 62% to 'Oceanomicrobium pacificus' KN286. Analyses of both 16S rRNA gene and genome sequences indicated that GXU_MW_L88T formed a unique branch within the Rhodobacterales. The genomic DNA G+C content of strain GXU_MW_L88T is 61.3%. The Q-10 was identified as the sole respiratory quinone, while C16 : 1 ω7c and iso-C10 : 03-OH constituted the predominant fatty acids (≥10%). The polar lipids were phosphatidylglycerol, an unidentified amino lipid, an unidentified aminophospholipid, two unidentified phospholipids and six unidentified lipids. Strain GXU_MW_L88T can produce l-hydroxocobalamin with a yield of up to 284 µg ml-1. Based on the above features, strain GXU_MW_L88T is suggested to represent a novel species within a novel genus under the order Rhodobacterales, for which the name Jujubicoccus corallicola gen. nov., sp. nov. is proposed, with the type strain GXU_MW_L88T (=KCTC 8863T=MCCC 1K09341T). The new family Agaricicolaceae fam. nov. is also proposed phylogenomically to accommodate the genera Agaricicola and 'Polymorphum'.
