Isolation of Native Soil Microorganisms with Potential for Breaking Down Biodegradable Plastic Mulch Films Used in Agriculture

Shinella agrisoli sp. nov., Isolated from a Pymetrozine-Degrading Bacterial Community

A Gram-stain negative, aerobic, motile, rod-shaped bacterium, designated BYT-45T, was isolated from a pymetrozine-degrading bacterial community. Strain BYT-45T was able to grow optimally in the presence of 0.5% (w/v) NaCl, at pH 7.0 and 30 °C. The 16 S rRNA gene sequence of strain BYT-45T showed the highest sequence similarity to Shinella pollutisoli AH-1T (98.1%). The average nucleotide identity and digital DNA-DNA hybridization values between BYT-45T and the type strains of the genus Shinella were 81.1-87.1% and 24.0-33.0% respectively. The draft genome sequence of BYT-45T was 5.46 Mb, with DNA G + C content of 66.0%. The major isoprenoid quinone was ubiquinone 10 and C19:0 cyclo ω8c, summed feature 8 (C18:1 ω6c and/or C18:1 ω7c), C16:0 and C18:1 ω7c 11-methyl constituted the major cellular fatty acids. The main polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylethanolamine, phosphatidylcholine, unidentified aminolipid, unidentified aminophospholipid, two unidentified phospholipids and two unidentified lipids. On the basis of evidence presented in this study, strain BYT-45T represents a novel species of the genus Shinella, for which the name Shinella agrisoli sp. nov. is proposed. The type strain is BYT-45T (= CCTCC AB 2024007T = KCTC 8545T).
