Self-labeling proteins (SLPs) enable the site-specific labeling of genetically expressed proteins with synthetic tags. BenzoTag was evolved from the commonly use self-labeling protein HaloTag7, for improved labeling kinetics with the fluorogenic dye, Bz1. This developed Bz1•BenzoTag conjugate was successfully applied in wash-free live cell imaging of intracellularly expressed BenzoTag and shown to have orthogonal reactivity against the HaloTag7•CA-JF635 system.
