Abstract:
Developing efficient coating strategies to address the low tolerance of microbes to acid stress remains challenging. Inspired by cryptobiosis in nature, we combined tannic acid (TA) and hydroxypropyl-β-cyclodextrin (HP-β-CD) and employed a one-step vortexing process to form a biocompatible multifunctional coating on the surface of Lactiplantibacillus plantarum XJ25 (LpX) for enhanced survival under acid stress. Molecular dynamics simulations revealed that noncovalent interactions guided the co-assembly of TA and HP-β-CD, enabling the resulting complex (HTc) to assemble stably on the bacterial cell membrane. After 3 h of acid stress at pH 2.5, coated LpX exhibited a survival rate 41.47 percentage points higher than that of native LpX. Proteomic analysis further suggested that the coating may promote the adaptation of LpX to acid stress. In practical wine malolactic fermentation (MLF), coated LpX maintained consistently high viable cell counts and completed malate metabolism 2 days earlier than LpX. This study demonstrates the advantages of HTc as a multifunctional coating material and highlightes its potential to enhance microbial adaptation to acid stress.
