Abstract:
Cancer initiation and progression are intricately linked with abnormal epigenetic modifications. The detection of these epigenetic alterations throughout tumorigenesis is essential for early cancer diagnosis and effective treatment. Cleavage Under Targets and Tagmentation (CUT&Tag) is a relatively new method for profiling diverse chromatin components. Here, we describe the genome-wide profiling of histone modifications by CUT&Tag in cancer tissues. Lightly cross-linked nuclei are immobilized on beads and incubated with a specific primary antibody against histone modifications. Then a secondary antibody is added, followed by protein AG-Tn5 transposase fusion protein. Tn5 transposase is activated to cleave the accessible chromatin proximal to antibody-bound target loci. PCR amplification is conducted to generate the next-generation sequencing (NGS) libraries.
