Abstract:
Lectins have the capacity to interact with cell surface carbohydrates, where they can influence various cellular processes. While early studies utilized lectin-induced cell agglutination to assess general binding characteristics, flow cytometry offers a more precise and quantitative method for measuring lectin interactions with specific cell populations. This technique is particularly valuable in studying galectins, a family of lectins that exhibit distinct binding preferences to red blood cells (RBCs) based on their blood type. This approach enables the determination of each lectin's binding specificity toward cell surface blood group antigens and other carbohydrate modifications without the potential confounding effects that differences in quaternary structure might have on their agglutination capabilities. We describe herein the production of fluorescently labeled galectins to analyze their specificity toward RBCs obtained from individuals with different blood types using flow cytometry.
