Abstract:
Glioblastoma (GBM) remains one of the most aggressive and heterogeneous brain tumors, with a high degree of molecular complexity that challenges both diagnosis and treatment. To address these challenges, this chapter details a methodology that utilizes the GeoMx Digital Spatial Profiler (GeoMx DSP, NanoString Technologies, Bruker) for comprehensive molecular profiling of GBM. The chapter outlines the application of GeoMx DSP in combination with the manufacturer's Whole Transcriptome Atlas (WTA) and Next-Generation Sequencing (NGS), enabling spatially resolved transcriptomic analysis within preserved tissue architecture. This integrated approach allows for detailed characterization of the molecular landscape in GBM at both spatial and transcriptional levels, facilitating the discovery of tumor microenvironment interactions, cellular heterogeneity, and potential therapeutic targets, thus advancing GBM research and clinical applications by supporting personalized treatment development.
