Abstract:
The respiratory epithelium (RE) and olfactory epithelium (OE) of the nasal mucosa are structurally similar yet functionally distinct: RE primarily facilitates air conditioning and host defense, while OE is responsible for odor detection and signal transmission. Although organoids have been applied in previous studies of both epithelia, there remains considerable room for refinement and further development. Here, we isolated primary cells from the respiratory and olfactory regions of the mouse nasal mucosa and established corresponding organoids using both three-dimensional (3D) and air-liquid interface (ALI) culture methods. The resulting in vitro models exhibited distinct lineage preferences and characteristics mirroring their tissue of origin. Furthermore, we demonstrated that IL-17a significantly inhibits ciliogenesis under the RE-ALI model, an effect that could be partially rescued by IL-17a inhibition.
