Background:
Bladder cancer (BC) is one of the most common malignant tumours of the urinary system and represents an important public health problem. Single nucleotide polymorphisms (SNPs) are common human genetic variations and may affect cancer risk by altering various genetic processes such as vitamin D metabolism and tumour suppression. CYP24A1 rs1570669 and rs2762934 polymorphisms may be relevant to cancer biology through their potential effects on the degradation of active vitamin D metabolites. These polymorphisms have been associated with the risk of urinary cancer, including BC, in an Asian population. To the best of our knowledge, the impact of CYP24A1 rs2762934 and rs1570669 variants on BC susceptibility has not yet been investigated in any Caucasian cohort. Consequently, this study represents the first attempt to evaluate their associations with BC risk within a Turkish population.
Methods And Results:
The present study investigated CYP24A1 gene polymorphisms in 152 BC patients and 178 healthy controls using a TaqMan allelic discrimination assay with real-time PCR (qPCR). Neither polymorphism was significantly associated with BC risk, although the A alleles of both variants showed a non-significant trend towards increased odds of BC. Additionally, both groups differed significantly regarding age, BMI, and smoking status, but not regarding other characteristics, such as vitamin D levels.
Conclusions:
The CYP24A1 rs2762934 and rs1570669 polymorphisms were not significantly associated with BC susceptibility in this Turkish cohort. Despite this, the A alleles of both variants showed a non-significant trend toward increased odds of BC, which may warrant further investigation in larger cohorts.
