Abstract:
P/Q-type voltage-dependent calcium channels (VDCCs) are involved in the regulation of neurotransmitter release and in the integration of dendritic inputs in the central nervous system (CNS). However, their precise localization and protein networks in the CNS remain elusive due to the lack of molecular tools applicable to the delicate and complex three-dimensional environment of the living brain. In this study, we synthesized peptide-based probes using ω-agatoxin IVA (AgTX), a specific channel blocker for P/Q-type VDCCs derived from the funnel-web spider Agelenopsis aperta. The far-red fluorophore-conjugated AgTX probe allowed the visualization of P/Q-type VDCCs in mouse cerebellum at subcellular spatial resolution by CLSM imaging. Moreover, by using a photosensitizer-conjugated AgTX, we successfully conducted photooxidation-driven proximity labeling of P/Q-type VDCCs in fixed mouse brain tissues.

Voltage-gated Ion Channels
Generally, all voltage-gated ion channels have a 'voltage-sensing domain' that spans the lipid bilayer. The charged residues in the sensor move in response to the membrane potential changes that open the channel allowing ions movement. There are several types of...
Mechanically-gated Ion Channels
Non-gated Ion Channels
Compared to the gated ion channels, the non-gated channels, also known as leakage or passive channels, have no gating mechanism.
Ion Channels
Ion channels are specialized integral membrane proteins on the plasma membrane that allow specific...

Three Subfamilies of Ligand-gated Ion Channels
Ligand-gated ion channels fall into three subfamilies. The 'Cys-loop' includes the nicotinic acetylcholine receptors, γ-aminobutyric acid (GABA), glycine, and 5-hydroxytryptamine receptors. The second one is the 'Pore-loop' channels that include the...
