Abstract:
Ulcerative colitis (UC) is a persistent inflammatory disease affecting the colorectum, for which no curative treatment is currently available. Elucidating the molecular mechanisms underlying UC pathogenesis may facilitate the development of novel therapeutic strategies. N6-methyladenosine (m6A) is a significant post-transcriptional RNA modification. YTH domain-containing family protein 2 (YTHDF2), a well-characterized m6A reader, plays a role in disease processes by facilitating the decay of target mRNAs. However, the specific role of YTHDF2 in UC remains largely unknown. In this study, bioinformatics analysis revealed elevated YTHDF2 expression in UC tissues, which was subsequently validated in lipopolysaccharide -stimulated Caco-2 and HT-29 cells as well as in dextran sulfate sodium-induced murine colitis. Knockdown of YTHDF2 alleviated pyroptosis and inflammatory responses both in vitro and in vivo, as revealed by reduced levels of NLRP3, ASC, cleaved caspase-1, and GSDMD-N, decreased caspase-1 activity, and diminished TNF-α, IL-6, IL-1β, and IL-18 secretion. These effects were mediated through suppression of the MAPK/NF-κB signaling axis. Mechanistically, YTHDF2 directly bound to and destabilized CCDC80 transcripts, leading to downregulation of CCDC80 and subsequent activation of MAPK/NF-κB signaling. Functional rescue experiments revealed that silencing CCDC80 partially reversed the anti-pyroptotic and anti-inflammatory effects of YTHDF2 knockdown. Collectively, our findings establish a critical role of YTHDF2 in UC pathogenesis and indicate its potential as a therapeutic target.

Inflammatory Bowel Disease I: Ulcerative Colitis
Inflammatory bowel disease, or IBD, encompasses a group of disorders characterized by chronic inflammation or ulceration of the gastrointestinal tract.
Risk Factors
The exact cause of IBD remains unclear, although it is believed to be due to a mix of genetic, environmental, microbial, and immune factors. Genetic factors are significant in determining susceptibility to IBD, with family history being a critical risk factor. Individuals with a first-degree relative who has IBD are at...

Inflammatory Bowel Disease II: Crohn's Disease
Inflammatory bowel disease, commonly known as IBD, refers to a collection of disorders that lead to persistent inflammation of the gastrointestinal tract. The two types of IBD are ulcerative colitis, which impacts the colon, and Crohn's disease, which can involve any part of the gastrointestinal segment.
Crohn's disease
Crohn's disease is a chronic, systemic inflammatory bowel disease (IBD) that predominantly affects the gastrointestinal tract. It is marked by transmural...
