Abstract:
The mechanistic basis of borrelidin biosynthesis in Streptomyces JK1-critical for antagonizing Pythium myriotylum-remains unclear. To identify the key regulatory genes, we generated a UV-induced mutant library and screened strains that lost borrelidin production and antifungal activity. A UV-induced mutant (60 s1) deficient in borrelidin production and inhibition was characterized, with whole-genome resequencing identifying LtrA as a key positive regulator. RT-qPCR validated that LtrA upregulates borB, an essential gene in borrelidin biosynthesis, establishing a core regulatory cascade. Molecular assays revealed LtrA interacts with FtsK/SpoIIIE (genome stability) and Crp/Fnr (cAMP signaling), integrating genomic integrity and signaling pathways. While cAMP rescued ltrA mutant morphology, it failed to restore the inhibitory activity of JK1 against Pythium myriotylum, indicating LtrA-mediated borrelidin regulation is distinct from morphological control. Overexpression of ltrA enhanced borrelidin production by 78.84%, confirming its regulatory centrality. ltrA overexpression boosts Streptomyces' biocontrol, offering a microbial engineering strategy for agricultural disease management.
