Abstract:
The core dilemma of senescence mitigation lies in the disrupted and even detrimental communication between senescent cells and local immune cells. Existing delivery methods cannot precisely establish protective intercellular communication channels across cell types within the enclosed lesion cavity. In this study, we utilize Chinese microneedle technology for precise, minimally invasive localization of aged lesions, and to release spatially targeted and temporally sustained dual-field to create intercellular communication channels between macrophages and senescent cells. The unique physical properties of this Chinese microneedle system confine the current path to specific spatiotemporal windows, releasing an electro-field while simultaneously activating dynamic van der Waals forces that release functionalized hydrogel microparticles, thereby superimposing a vita-field. Within the dual-field active zone, the electro-field not only attenuates cellular senescence but also forms a communication channel from senescent cells to macrophages, driving macrophages into a hyperactivated state. Meanwhile, the vita-field stabilizes the macrophage membrane, enabling long-term, stable communication between hyperactivated macrophages and senescent cells. Ultimately, the intercellular communication channels established by this Chinese microneedle system alleviate cellular senescence and apoptosis, supporting the repair of aged lesions.
