Abstract:
Zebrafish have emerged as a powerful model to study myelination and oligodendrocyte dynamics due to their optical transparency and regenerative capacity. This chapter provides a comprehensive set of protocols for analyzing oligodendrocytes and myelination in zebrafish embryos and larvae and also adults. We describe step-by-step methods for live imaging using transgenic lines, whole-mount and sectioned immunohistochemistry, and Western blot analysis. These approaches allow detailed spatial and temporal resolution of oligodendrocyte markers in vivo, ex vivo, and in vitro, facilitating the investigation of developmental myelination and remyelination following injury.
