Mutant of phi X174 accessible to host-controlled modification
Journal of Virology
|May 1, 1969
Abstract:
A proflavine-induced phiX174 mutant was isolated, phiX174 sB1, which is accessible to restriction and modification by B-host specificity, but not by K-host specificity.
Insights
A new mutant of bacteriophage phiX174, named phiX174 sB1, was created. This mutant can be restricted and modified by B-host specificity but not K-host specificity.
Area of Science:
- Molecular biology
- Virology
- Bacteriophage genetics
Background:
- Bacteriophage phiX174 is a well-studied model organism in molecular biology.
- B-host and K-host specificity are important factors in bacteriophage-host interactions and replication.
Purpose of the Study:
- To isolate and characterize a mutant of bacteriophage phiX174 with altered host specificity.
- To investigate the genetic basis of host specificity in bacteriophages.
Main Methods:
- Induction of mutations in bacteriophage phiX174 using proflavine.
- Isolation and selection of mutant bacteriophages.
- Testing of mutant bacteriophage susceptibility to restriction and modification by different host specificities (B-host and K-host).
Main Results:
- A proflavine-induced mutant, designated phiX174 sB1, was successfully isolated.
- The phiX174 sB1 mutant demonstrated accessibility to restriction and modification by B-host specificity.
- The phiX174 sB1 mutant was not accessible to restriction and modification by K-host specificity.
Conclusions:
- The isolated phiX174 sB1 mutant exhibits a specific alteration in its host interaction profile.
- This finding contributes to understanding the genetic determinants of bacteriophage host specificity.
