    Murine intracisternal A particles possess RNA-dependent DNA polymerase and polyadenylic acid-containing RNA. These A-particles are not closely related genetically to murine type C viruses, based on DNA transcript hybridization studies.

    Background:

    • Intracisternal A particles (A-particles) are endogenous retroviral elements found in mice.
    • These particles contain RNA-dependent DNA polymerase activity and high molecular-weight RNA with polyadenylic acid (poly[A]) tails.

    Purpose of the Study:

    • To investigate the relationship between murine intracisternal A particles and murine type C viruses.
    • To characterize the endogenous reverse transcriptase activity and RNA content of A-particles.

    Main Methods:

    • Isolation of A-particle preparations from murine neuroblastoma (N18) and plasmacytoma (MOPC-104E) cell lines.
    • Assay of RNA-dependent DNA polymerase activity using various oligo- and polyribonucleotides.
    • Molecular hybridization experiments comparing A-particle RNA and DNA transcripts with type C viral RNA.

    • An alternative explanation suggests A-particle DNA transcripts may originate from a small, unrelated portion of the genome.
