    Background:

    • Polycythemia, an elevated red blood cell count, can be induced by various methods, including red blood cell (RBC) transfusions.
    • Thrombocytopenia, a low platelet count, has been observed in polycythemic states, but the underlying mechanisms require clarification.

    Purpose of the Study:

    • To investigate the relationship between transfusion-induced polycythemia and thrombocytopenia in mice.
    • To determine if decreased platelet production, shortened platelet survival, or splenic sequestration contribute to thrombocytopenia in this model.

    Main Methods:

    • Polycythemia was induced in mice via weekly injections of washed packed RBCs.
    • Platelet production was assessed using 75SeM incorporation.
    • Platelet survival was evaluated using 51Cr-labeled platelets.
    • Splenic platelet sequestration was measured by 51Cr-bound radioactivity.

    • This mechanism differs from that observed in hypoxia-induced polycythemia.
    • Expanded blood volume is the key factor contributing to thrombocytopenia in this experimental model.
