Type I and Type II interferon receptors.

P Orchansky, D Novick, D G Fischer

    Journal of Interferon Research
    |January 1, 1984
    PubMed
    Summary

Interferon (IFN)-gamma uniquely binds to its specific receptor, distinct from receptors for IFN-alpha and IFN-beta. This research differentiates Type I and Type II interferon receptors based on binding specificity.

    Area of Science:

    • Immunology
    • Cell Biology
    • Virology

    Background:

    • Interferons (IFNs) are crucial cytokines involved in immune responses.
    • Different types of interferons (IFN-alpha, IFN-beta, IFN-gamma) exist, but their receptor specificities require clear definition.

    Purpose of the Study:

    • To investigate and differentiate the specific binding sites for IFN-gamma compared to IFN-alpha and IFN-beta.

    Main Methods:

    • Binding site competition assays using radiolabeled IFN-gamma (125I-IFN-gamma).
    • Utilized homogeneous preparations of IFN-alpha 2, IFN-beta, and IFN-gamma.
    • Experiments conducted on WISH and FS11 cell lines.

    Main Results:

    • Only IFN-gamma effectively competed for the 125I-IFN-gamma binding site.
    • IFN-alpha and IFN-beta did not compete for the IFN-gamma specific binding site.
    • Demonstrated distinct cellular receptors for IFN-gamma versus IFN-alpha/beta.

    Conclusions:

    • A clear distinction between IFN-gamma receptors and IFN-alpha/beta receptors was established.
    • Proposed naming the IFN-alpha/beta receptor as "Type I" and the IFN-gamma receptor as "Type II".
