Enzymehistochemical staining of T mu lymphocytes for glycyl-proline-4-methoxy-beta-naphthylamide-peptidase (DAP IV)

|June 1, 1982
Abstract:
Human T mu lymphocytes are detected in a histochemical reaction with glycyl-proline-4-methoxy-beta-naphthylamide-peptidase (DAP IV; EC 3.4.14.4). DAP IV occurs in 59.4 +/- 8.25 of human T lymphocytes in blood and marrow. Direct biochemical assays were in accord with the cytochemical findings. DAP IV-positive lymphocytes accounted for 89.5 +/- 0.6% of T-lymphocytes bearing Fc receptor for IgG contained 11.3 +/- 8.3% T-lymphocytes bearing Fc receptor for IgM.
Insights
Dipeptidyl peptidase IV (DAP IV) is present in human T lymphocytes. This enzyme was detected in over 59% of T cells in blood and marrow samples.
Area of Science:
- Immunology
- Biochemistry
Background:
- Human T lymphocytes play crucial roles in immune responses.
- Dipeptidyl peptidase IV (DAP IV) is an enzyme involved in various biological processes.
Purpose of the Study:
- To investigate the presence and distribution of DAP IV in human T lymphocytes.
- To correlate DAP IV expression with T lymphocyte subsets.
Main Methods:
- Histochemical staining using glycyl-proline-4-methoxy-beta-naphthylamide-peptidase.
- Direct biochemical assays.
- Flow cytometry to identify T-lymphocytes bearing Fc receptors for IgG and IgM.
Main Results:
- DAP IV was detected in 59.4 +/- 8.25% of human T lymphocytes in blood and marrow.
- Biochemical assays confirmed cytochemical findings.
- DAP IV-positive lymphocytes constituted 89.5 +/- 0.6% of T-lymphocytes with Fc receptors for IgG and 11.3 +/- 8.3% with Fc receptors for IgM.
Conclusions:
- DAP IV is a significant marker for human T lymphocytes.
- The study provides quantitative data on DAP IV expression in T cell populations.
