[Mezlocillin concentrations in human lung tissue].

U Kroening, S Liebig, M Wundschock

    Summary

Mezlocillin concentrations in lung and muscle tissue were evaluated post-pneumonectomy. Lung tissue concentrations exceeded minimum inhibitory concentrations for most bacteria for two hours, suggesting effective antibacterial activity.

    Area of Science:

    • Pharmacokinetics and Pharmacodynamics
    • Antimicrobial Therapy
    • Surgical Infections

    Context:

    • Assessing antibiotic efficacy in surgical patients.
    • Evaluating drug distribution in specific tissues post-pneumonectomy.

    Purpose:

    • To determine mezlocillin concentrations in serum, lung parenchyma, and skeletal muscle.
    • To compare these concentrations against minimum inhibitory concentrations (MICs) for bacterial pathogens.

    Summary:

    • Antibiotic concentrations of mezlocillin were measured in serum, lung, and muscle tissues of 28 patients following pneumonectomy.
    • Intravenous bolus injection of 5g mezlocillin was administered 30-150 minutes prior to sampling.
    • Lung tissue concentrations remained above MICs for most bacteria for 2 hours; muscle tissue concentrations were also found to be effective.

    Impact:

    • Provides crucial data for optimizing mezlocillin dosing in post-pneumonectomy patients.
    • Highlights the need for higher doses in cases of less sensitive bacterial strains.
    • Informs clinical decisions regarding antibiotic selection and dosage in thoracic surgery.
