Increased thymic hormone responsive suppressor T lymphocyte function in chronic active hepatitis.

    Summary

Chronic active hepatitis (CAH) patients exhibit reduced suppressor T lymphocyte function. Thymosin fraction 5 treatment in vitro restored this function, indicating potential therapeutic benefits for immune modulation in CAH.

    Area of Science:

    • Immunology
    • Hepatology
    • T cell biology

    Background:

    • Chronic active hepatitis (CAH) is associated with immune dysregulation.
    • Suppressor T lymphocyte function plays a critical role in maintaining immune homeostasis.
    • Impaired T cell function may contribute to the pathogenesis of CAH.

    Purpose of the Study:

    • To evaluate mitogen-induced suppressor T lymphocyte function in patients with CAH.
    • To assess the in vitro effect of thymosin fraction 5 on the suppressive activity of peripheral blood mononuclear cells (PBM).

    Main Methods:

    • Peripheral blood mononuclear cells (PBM) were isolated from patients with CAH and healthy controls.
    • Suppressor T cell activity was measured using mitogen-induced assays.
    • The effect of thymosin fraction 5 on PBM was evaluated in vitro, both with and without mitogen stimulation.

    Related Experiment Videos

    Main Results:

    • Suppressor T cell activity was significantly decreased in CAH patients compared to controls (P < 0.001).
    • Thymosin fraction 5 enhanced helper effects in both patients and controls in the absence of mitogens.
    • Mitogen-activated, thymosin-treated PBM from CAH patients showed a marked increase in suppressor activity (P < 0.001), unlike controls.

    Conclusions:

    • Thymosin fraction 5, containing polypeptides, can exert a helper effect on lymphocytes.
    • PBM from CAH patients harbor a subset of lymphocytes capable of expressing suppressive function after thymosin treatment.
    • Thymosin fraction 5 demonstrates potential for in vitro restoration of suppressor T cell function in patients with CAH.
