    Background:

    • Huntington's disease (HD) is a neurodegenerative disorder.
    • Cellular abnormalities are implicated in HD pathogenesis.
    • The nucleus accumbens, caudate nucleus, and frontal cortex are key brain regions affected in HD.

    Purpose of the Study:

    • To investigate ultrastructural changes in specific brain regions of patients with Huntington's disease.
    • To identify the presence and frequency of nuclear membrane indentations (NMI) in neurons from affected individuals.

    Main Methods:

    • Electron-microscopical examination was conducted on post-mortem brain tissue.
    • Samples were obtained from 21 patients diagnosed with Huntington's disease.
    • Tissue from 21 age-matched healthy individuals served as controls.

    Main Results:

    • Nuclear membrane indentations (NMI) were observed in 25% of nucleus accumbens neurons, 29% of caudate nucleus neurons, and 20% of frontal cortex neurons in HD patients.
    • NMI were significantly less frequent in control subjects (4% in nucleus accumbens, 4% in caudate nucleus, 38% in frontal cortex).
    • The prevalence of NMI differed across brain regions in both HD patients and controls.

    Conclusions:

    • The increased prevalence of NMI in specific brain regions of HD patients suggests a potential role in the disease's cellular pathology.
    • Further research is warranted to elucidate the pathogenetic mechanisms underlying NMI formation in Huntington's disease.
    • NMI may represent a novel cellular marker for neurodegeneration in HD.
